The global spread of antibiotic resistance is regarded as one of the most significant health threats to society. While the development of new antibiotics is crucial, there is also an urgent need for next-generation analytical methods to monitor the physiological state of live bacteria within heterogeneous populations and their responses to environmental stress. In this study, we present the development of a Nile Red–based solvatochromic antimicrobial peptide, UNR-1. We demonstrated that UNR-1 selectively stained the cell envelopes of both Gram-positive and Gram-negative bacteria, showing high selectivity over eukaryotic cells. The solvatochromic fluorescence of UNR-1 was responsive to perturbations in the bacterial cell envelope induced by external physical and chemical stressors. To enable an analysis of the perturbations at the level of cell envelope in individual cells, we established a robust protocol for calculating the generalised polarisation of fluorescence, adapted for flow cytometry. This methodology facilitated the rapid detection of cell envelope perturbations caused by exposure to antibiotics, heat shock, and other stress factors. ⸻
